Dr Kenneth Fahy, SiriusXT, presented at the EMBO workshop that took place in EMBL, covering the topic “Cryo Soft X-ray Tomography for efficient targeting and volume context in CLEM and Volume EM correlative workflows”.
Soft X-ray tomography (SXT) is a powerful standalone imaging technique with the unique capability to non-destructively image the whole 3D subcellular structures of intact hydrated biological cells or tissue samples with nanometre resolution.
SXT is also extremely beneficial when incorporated into correlative imaging workflows, by:
⭐ Identifying regions of interest (RoI) within large samples, providing precise coordinates for lamella production and subsequent TEM imaging.
⭐ Providing structural context to help analyse results by imaging a larger volume around the region of interest.
⭐ Increasing imaging through-put.
⭐ Using non-labour intensive sample preparation techniques.
⭐ Providing the ability to track and validate the delivery of nanoparticles to targeted endpoints in cells.
