The SXT-100 contains an integrated 5-channel epi-fluorescent microscope for cell mapping and correlative microscopy.
Correlative FM-SXT imaging of cells incubated with nanoparticles.
SXT-100 data reveals branched and folded mitochondria in the vicinity of the cell nucleus.
Fluorescence image of mCherry labelled Hepatitis C viral replicon and Mitotracker green labelled mitochondria overlaid with soft X-ray projection and a reconstruction slice through the infected HUH 7 cell.
Samples of C. elegans larvae of strain DM8005 were high pressure frozen using the waffle method. Multiple 5 micron thick slabs spanning an L2 larva were extracted using the Serial Lift-Out technique, and placed on rectangular mesh grids. Soft X-ray tomography was performed on individual slabs using the SXT-100 soft x-ray microscope at SiriusXT. The data reveals internal structures of fully hydrated C. elegans tissue.
SXT reconstruction of the mid-body of C. elegans.
Cryo soft X-ray tomography is an important tool to improve information content and targeting efficiency in in situ structural biology workflows.
Downstream cryo-ET data from SXT-exposed samples showed no devitrification artifacts and the resulting tomograms are morphologically indistinct from non-exposed areas of the sample.
Longitudinal Lift-Outs of C. elegans anterior region, several microns thick.
High contrast SXT data from Euglena, combined with chlorophyll autofluorescence.
